چکیده
s | 19 Methods: We studied 20 healthy volunteers who were tested on two occasions receiving either ebselen (3600mg over 24 hours) or identical placebo in a double-blind, random-order, cross-over design. Two hours after the final dose of ebselen/placebo, participants underwent proton magnetic resonance spectroscopy (1H MRS) at 7 tesla (7T) with voxels placed in anterior cingulate and occipital cortex (Figure 1). Neurometabolite levels were calculated using an unsuppressed water signal as a reference and corrected for individual cerebrospinal fluid content in the voxel. Results: Ebselen produced no effect on neurometabolite levels in the occipital cortex. In the anterior cingulate cortex, ebselen lowered concentrations of inositol as well as those of glutathione, glutamine, glutamate and Glx (Table 1). Conclusions: The study suggests that at the dosage used, ebselen produces a functional inhibition of IMPase in the human brain. The ability of ebselen to lower indices of glutamate activity are consistent with its action, reported in animal experimental work, to inhibit the enzyme, glutaminase. Ebselen appears to have potential as a repurposed treatment for bipolar disorder and it would be of interest to see if similar biochemical alterations are produced by ebselen treatment in this patient group. Antidepressant Treatment for Acute Bipolar Depression: An Update
While studies in the past have focused more on treatment of the manic phase of bipolar disorder (BD), recent findings demonstrate the depressive phase to be at least as debilitating. However, in contrast to unipolar depression, depression in bipolar patients exhibits a varying response to antidepressants, raising questions regarding their efficacy and tolerability. Methods. We conducted a MEDLI...

Although rarely reported, the induction of hypomanic/manic episodes due to sudden or gradual cessation of antidepressant drugs is a phenomenon observed in clinical settings. Herein we present 2 patients that had manic episodes induced by gradual cessation of antidepressant drugs. Common features of both cases were as follows: patients were female; a major depressive episode was the reason for s...

Sociodemographic and clinical features of antidepressant-induced hypomanic and manic switch in patients with bipolar disorder Objective: The aim of this study was to identify sociodemographic, familial, childhood and various clinical characteristics of bipolar disorder patients in whom hypomanic/manic switches had been observed during treatment with antidepressant drugs. Methods: One hundred si...
متن کاملHypomanic shift observed during rTMS treatment of patients with unipolar depressive disorder: four case reports
OBJECTIVE Repetitive transcranial magnetic stimulation (rTMS) can enhance the excitement of the brain through adjusting the biological activities of the cerebral cortex and has wide biological effects, making it one basic mechanism of therapy for depression. In the treatment of unipolar depressive disorder, almost in every treatment method, hypomanic and manic shifts can be observed. There is s...

OBJECTIVES Bipolar type II (BP II) disorder is thought to be distinct from BP I disorder on genetic and biological grounds, and it is not merely a milder form of the illness. It affects 1.5-2.5% of the US adult population, and is characterized by highly recurrent depressive episodes with a substantial morbidity from alcoholism and non-affective psychopathology, and a higher suicide rate than ei...
